A spatiotemporal study of bacterial community profiles associated with Atlantic bluefin tuna larvae, Thunnus thynnus L., in three Mediterranean hatcheries ArchiMer
Gatesoupe, Joel; Coves, Denis; Ortega, Aurelio; Papandroulakis, Nikos; Vadstein, Olav; De La Gandara, Fernando.
During the first 2 years of larval rearing trials with Atlantic bluefin tuna, survival was a challenging issue. As bacterial colonization of the gut has been shown to play a key role for other species, we studied the profiles of the microbiota associated with individual larvae at different stages in three distant hatcheries. The Bacterial Community Profile (BCP) was quantified based on PCR-DGGE analyses of partial amplicons from 16S rDNA. Considerable individual variability in BCP was observed before onset of feeding, and the BCP did not show regular fluctuation during ontogenesis. Microalgae were added to the rearing tanks in two of the three hatcheries, but it was not possible to distinguish the effect of location from the effect of algal addition on...

Comparison of the effects of the dietary addition of two lactic acid bacteria on the development and conformation of sea bass larvae, Dicentrarchus labrax, and the influence on associated microbiota ArchiMer
Lamari, Faouzi; Castex, Mathieu; Larcher, Thibaut; Ledevin, Mireille; Mazurais, David; Bakhrouf, Amina; Gatesoupe, Joel.
Probiotics may have many effects on health and development of fish larvae. One of the most promising is related to spinal conformation, though the mode of action is not clearly understood. The present study attempted to investigate the effects of two strains of lactic acid bacteria on associated microbiota, histological development and gene expression. Sea bass larvae were fed since 5 dph (day post hatch) with either a standard control diet (Diet C), or the same diet supplemented with Pediococcus acidilactici MA18/5M (Diet P), or with an autochthonous strain of Lactobacillus casei (X2; Diet L). The two lactic acid bacteria were incorporated in the diets at the levels of 106 and 107 CFU (colony forming units) g-1 in two consecutive experiments,...

Cross effects of the strain of dietary Saccharomyces cerevisiae and rearing conditions on the onset of intestinal microbiota and digestive enzymes in rainbow trout, Onchorhynchus mykiss, fry ArchiMer
Wache, Yann; Auffray, Francoise; Gatesoupe, Joel; Zambonino, Jose-luis; Gayet, Vincent; Labbe, Laurent; Quentel, C.
Two strains of Saccharomyces cerevisiae were tested as probiotics for rainbow trout fry, during the first month of feeding. Each strain was introduced into separate diets, at the rate of 106 CFU g− 1 and their effects were compared with those of a control diet. Two rearing conditions were simultaneously compared, to test the adaptability of the probiotic treatment. From start feeding onwards, the water supply came from either spring or river, resulting in two different temperature ranges, 11¿11.5 and 7¿8 °C respectively. Growth and development were optimal in spring water, while some delay was observed with colder river water. A slight but significant increase in mortality was also observed in the river group. In all groups, the counts of...

Effect of dietary inulin and oligosaccharides as prebiotics for weaning turbot, Psetta maxima (Linnaeus, C. 1758) ArchiMer
Mahious, A.s.; Gatesoupe, Joel; Hervy, Magali; Metailler, Robert; Ollevier, F.
Preliminary experiments were undertaken to investigate the effect of dietary inulin (Raftiline ST), oligofructose (Raftilose P95) and lactosucrose on the growth and intestinal bacteria of the marine carnivorous turbot, Psetta maxima. Turbot larvae were weaned on compound diets containing 2% Rafiline ST, 2% Raftilose P95 or 2% lactosucrose; 2% cellulose was the carbon source in the control group. The final mean weight of the group weaned with Raftilose P95 was significantly higher than those observed with the other diets. The bacterial load was highly variable in weaning turbot, especially with respect to the putative Vibrio spp. growing on TCBS agar which, in general, seemed to be dominant. Of the total load of bacterial isolates from turbot weaned on...

Effects of different dietary levels of fish protein hydrolysates on growth, digestive enzymes, gut microbiota, and resistance to Vibrio anguillarum in European sea bass (Dicentrarchus labrax) larvae ArchiMer
Kotzamanis, Y. P.; Gisbert, E.; Gatesoupe, Joel; Zambonino, Jose-luis; Cahu, Chantal.
Two Fish Protein Hydrolysates (FPH) were incorporated into four diets prepared for start-feeding sea bass larvae, at two different levels (10% and 19% of total ingredients): a commercial FPH, CPSP, in which the molecular weight of the main fraction of soluble peptides (51%) was between 500-2500 Da, and an experimental FPH obtained by acidic silage of sardine offal, SH, with a main portion of soluble peptides (54%) ranging from 200 to 500 Da. The diet with 10% of the commercial FPH gave the best results in terms of growth, survival and intestinal development, as evaluated by the early activity of digestive enzymes in the brush border membrane (alkaline phosphatase and aminopeptidase N). This was related to the low level of Vibrio spp. counted in the larvae...

Influence of dietary live yeast on European sea bass (Dicentrarchus labrax) larval development ArchiMer
Tovar Ramirez, Dariel; Zambonino-infante, Jose-luis; Cahu, Chantal; Gatesoupe, Joel; Vazquez Juarez, R.
Recent results have demonstrated that live yeast in diets improved gut maturation in sea bass larvae. In this study, we tested the absence and two levels of live yeast in sea bass larvae diets. Specimens were fed from time of mouth opening to 37 days after hatching a diet of 0%, 1.1%, or 5.7% wet weight of live yeast (Debaryomyces hansenii CBS 8339). Yeast incorporation improved survival 10%, and reduced malformed larvae. In groups fed 1.1% yeast, only 1% of larvae were malformed, compared to 14% in the control group. Final mean weight in groups fed 1.1% yeast was twice that of other groups. Activities and concentrations of mRNA trypsin and lipase were higher in the two groups fed yeast than in the control group, whereas activity and concentration of mRNA...

Live yeasts in the gut: Natural occurrence, dietary introduction, and their effects on fish health and development ArchiMer
Gatesoupe, Joel.
This minireview summarizes the present state of knowledge concerning the importance of yeasts in fish gut. Yeasts have been commonly isolated in the gastrointestinal tract, and high population densities were sometimes noted in healthy fish, but the data were quite variable in terms of colony counts and taxonomical diversity. Rhodotorula sp. seemed relatively frequent in both marine and freshwater fish, and Debaryomyces hansenii has been found to be dominant in rainbow trout. Some other dominant strains have been described, such as Metschnikowia zobelii, Trichosporon cutaneum, and Candida tropicalis in marine fish, and Candida sp., Saccharomyces cerevisiae, and Leucosporidium sp. in rainbow trout. The natural proliferation of yeasts in fish mucus may be...

For most marine aquaculture species, one of the main bottlenecks is the stable production of high quality juveniles. The high and unpredictable mortality in the first weeks after hatching of marine fish larvae remains a challenging problem that needs to be solved. The severity of the problem differs between species, but cannot be considered adequately solved for any species. Both scientific evidence and experience in hatcheries for a variety of fish, shrimp and shellfish species are accumulating as support for the hypothesis that detrimental fish-microbe interactions are the cause of these problems. Host-microbe interactions in reared fish are still poorly understood, except for a few pathogens, and empirical data of the quality required to test this...

Ontogenèse, Développement et Physiologie chez les larves de poissons ArchiMer
Gatesoupe, Joel; Zambonino-infante, Jose-luis; Cahu, Chantal; Bergot, P.

Probiotic and formaldehyde treatments of Artemia nauplii as food for larval pollack, Pollachius pollachius ArchiMer
Gatesoupe, Joel.
Formaldehyde was used to disinfect Artemia cysts and nauplii, while introducing two probiotics in the enrichment process: Bactocell (Pediococcus acidilactici) and Levucell (Saccharomyces cerevisiae). The disinfectant was selected due to its potential compatibility with probiotics, since it was more effective against Gram-negative bacteria than against lactic acid bacteria and yeast. However, the presence of formaldehyde reduced the intake of P. acidilactici in Artemia. Consequently, the disinfection was stopped before Bactocell supplementation to the nauplii fed to pollack (Pollachius pollachius) larvae. The mean weight of pollack was higher with this probiotic treatment. Growth was even better with the combination of Levucell and Bactocell, but the yeast...

Probiotics and prebiotics for fish culture, at the parting of the ways ArchiMer
Gatesoupe, Joel.
After more than 20 years of research on probiotics for aquaculture, the treatment has become a reality in farms and hatcheries, mainly in those devoted to shrimp production. New insights into gut microbiota have recently offered the opportunity to reconsider the application to fish culture, still lagging behind shrimp industry in the use of probiotics.this regard. Additionally, the concept of prebiotics is emerging as another effective way of disease control in aquaculture. Prebiotics are food ingredients that beneficially affect the host by selectively stimulating the growth of and/or activating the metabolism of one or a limited number of health-promoting bacteria in the intestinal tract. While dietary applications of prebiotics and probiotics may be...

Selection of lactic acid bacteria as candidate probiotics and in vivo test on Artemia nauplii ArchiMer
Lamari, Faouzi; Sadok, Khouadja; Bakhrouf, Amina; Gatesoupe, Joel.
Lactic acid bacteria (LAB) were isolated from rotifer cultures in a marine hatchery to search for potential probiotics for marine animals. Fifteen strains were first selected among a total of 55, according to antibacterial activity against Vibrio sp. Among eight strains identified as Lactobacillus casei, four were highly adhesive, suggesting some ability for surface colonization. The other strains were identified as Lactobacillus plantarum, Lactobacillus dextrinicus, and Leuconostoc sp. To validate probiotic potential, Artemia were challenged against pathogenic Vibrio alginolyticus, with or without one of six selected LAB strains. The six strains protected Artemia against the pathogen, to some extent on condition that nutrient enrichment was provided. La....

The effects of dietary carbohydrate sources and forms on metabolic response and intestinal microbiota in sea bass juveniles, Dicentrarchus labrax ArchiMer
Gatesoupe, Joel; Huelvan, Christine; Le Bayon, Nicolas; Severe, Armelle; Aasen, Inga Marie; Degnes, Kristin F.; Mazurais, David; Panserat, Stephane; Zambonino, Jose-luis; Kaushik, Sadasivam.
The aim of this work was to investigate the catabolic process of three kinds of dietary carbohydrates in the gut of sea bass juveniles, with the possible contribution of the intestinal microbiota to the nutrition of the host, and the subsequent effects on intermediary metabolism. A first diet contained waxy maize (99% amylopectin), a highly digestible form of starch. A second diet was less quickly digestible due to its high amylose content of resistant starch. Two other diets contained fibre instead of starch, either only cellulose as control, or also other non-starch polysaccharides brought by lupin meal. The effect of the diets on the host confirmed previous results, with the stimulation of glucose storage in the liver in sea bass fed the starchy diets,...

Trial of probiotics to prevent the vertebral column compression syndrome in rainbow trout (Oncorhynchus mykiss Walbaum) ArchiMer
Aubin, Joel; Gatesoupe, Joel; Labbe, Laurent; Lebrun, Luc.
Two probiotics were tested as alternative treatment to limit the prevalence of the vertebral column compression syndrome (VCCS) in rainbow trout, compared with a preventive treatment with florfenicol. Either the antibiotic, or a lactic acid bacterium, Pediococcus acidilactici, or a yeast, Saccharomyces cerevisiae var. boulardii, was introduced into experimental diets, which were compared with the control diet without supplementation. The antibiotic caused some mortality during the first week after treatment, likely because of the toxicity of the compound. However, this antibiotic treatment limited the occurrence of VCCS to 3%, vs. 13% of fish affected in the control group. Pediococcus acidilactici provided the same level of protection as the antibiotic,...

Updating the importance of lactic acid bacteria in fish farming: natural occurrence and probiotic treatments ArchiMer
Gatesoupe, Joel.
Many recent papers have deepened the state of knowledge about lactic acid bacteria (LAB) in fish gut. In spite of high variability in fish microbiota, LAB are sometimes abundant in the intestine, notably in freshwater fish. Several strains of Streptococcus are pathogenic to fish. S. iniae and Lactococcus garvieae are major fish pathogens, against which commercial vaccines are available. Fortunately, most LAB are harmless, and some strains have been reported for beneficent effects on fish health. A major step forward of the recent years was those converging evidences that LAB can stimulate the immune system in fish. An open question is whether viability can affect immunostimulation. The issue is crucial to commercialise live probiotics rather than...
